Catheter-related infections in a northwestern São Paulo reference unit for burned patients care BJID
Campos Júnior,Cláudio Penido; Sanches,Patrícia; Tedokon,Elizabete Aparecida; Souza,Ana Carolina Remondi; Machado,Ricardo Luiz Dantas; Rossit,Andréa Regina Baptista.
Despite improvements in care and rehabilitation of burned patients, infections still remain the main complication and death cause. Catheter-related infections are among the four most common infections and are associated with skin damage and insertion site colonization. There are few studies evaluating this kind of infection worldwide in this special group of patients. Padre Albino Hospital Burn Care Unit (PAHBCU) is the only reference center in the Northwestern São Paulo for treatment of burned patients. This paper presents the results of a retrospective study aiming at describing the epidemiological and clinical features of catheter-related infections at PAHBCU.

Characterisation of volatile compounds produced by bacteria isolated from the spoilage flora of cold-smoked salmon ArchiMer
Joffraud, Jean-jacques; Leroi, Francoise; Roy, Caroline; Berdague, J.
This study investigated the volatile compounds produced by bacteria belonging to nine different bacterial groups: Lactobacillus sake, L. farciminis, L. alimentarius. Carnobacterium piscicola, Aeromonas sp., Shewanella putefaciens. Brochothrix thermosphacta, Photobacterium phosphoreum and Enterobacteriaceae isolated from cold-smoked salmon. Each bacterial group was represented by several strains. In addition, combinations of the groups were examined as well. Sterile blocks of cold-smoked salmon were inoculated, vacuum-packed and stored at 6 degreesC. After 40 days of storage at 6 degreesC, aerobic viable count and pH were recorded, the volatile fraction of the samples was analysed by gas chromatography-mass spectrometry (GC-MS). and spoilage was assessed by...

Characterization of a small cryptic plasmid from endophytic Pantoea agglomerans and its use in the construction of an expression vector Genet. Mol. Biol.
Rudi Emerson de Lima,Procópio; Araújo,Welington Luiz; Andreote,Fernando Dini; Azevedo,João Lúcio.
A circular cryptic plasmid named pPAGA (2,734 bp) was isolated from Pantoea agglomerans strain EGE6 (an endophytic bacterial isolate from eucalyptus). Sequence analysis revealed that the plasmid has a G+C content of 51% and contains four potential ORFs, 238(A), 250(B), 131(C), and 129(D) amino acids in length without homology to known proteins. The shuttle vector pLGM1 was constructed by combining the pPAGA plasmid with pGFPmut3.0 (which harbors a gene encoding green fluorescent protein, GFP), and the resulting construct was used to over-express GFP in E. coli and P. agglomerans cells. GFP production was used to monitor the colonization of strain EGE6gfp in various plant tissues by fluorescence microscopy. Analysis of EGE6gfp colonization showed that 14...

Characterization of dioxygenases and biosurfactants produced by crude oil degrading soil bacteria BJM
Muthukamalam,Santhakumar; Sivagangavathi,Sivalingam; Dhrishya,Dharmapal; Sudha Rani,Sadras.
ABSTRACT Role of microbes in bioremediation of oil spills has become inevitable owing to their eco friendly nature. This study focused on the isolation and characterization of bacterial strains with superior oil degrading potential from crude-oil contaminated soil. Three such bacterial strains were selected and subsequently identified by 16S rRNA gene sequence analysis as Corynebacterium aurimucosum, Acinetobacter baumannii and Microbacterium hydrocarbonoxydans respectively. The specific activity of catechol 1,2 dioxygenase (C12O) and catechol 2,3 dioxygenase (C23O) was determined in these three strains wherein the activity of C12O was more than that of C23O. Among the three strains, Microbacterium hydrocarbonoxydans exhibited superior crude oil degrading...

Chlorine dioxide against bacteria and yeasts from the alcoholic fermentation BJM
Meneghin,Silvana Perissatto; Reis,Fabricia Cristina; Almeida,Paulo Garcia de; Ceccato-Antonini,Sandra Regina.
The ethanol production in Brazil is carried out by fed-batch or continuous process with cell recycle, in such way that bacterial contaminants are also recycled and may be troublesome due to the substrate competition. Addition of sulphuric acid when inoculum cells are washed can control the bacterial growth or alternatively biocides are used. This work aimed to verify the effect of chlorine dioxide, a well-known biocide for bacterial decontamination of water and equipments, against contaminant bacteria (Bacillus subtilis, Lactobacillus plantarum, Lactobacillus fermentum and Leuconostoc mesenteroides) from alcoholic fermentation, through the method of minimum inhibitory concentration (MIC), as well as its effect on the industrial yeast inoculum. Lower MIC...

Cloning and characterization of the gene encoding the PepF endopeptidase from the aquatic bacterium Caulobacter crescentus BJM
Braz,Vânia S.; Lang,Elza A.S.; Marques,Marilis V..
The metallopeptidases have a very important role in bacteria, being involved in several processes that rely on protein turnover, such as nutrition, degradation of signal peptides, protein localization and virulence. We have cloned and characterized the gene of the metalloendopeptidase PepF from the aquatic bacterium Caulobacter crescentus. The gene upstream of pepF (orf1) encodes a conserved hypothetical protein found in Mycobacterium and Streptomyces. pepF is co-transcribed with the gene downstream (orf3), which encodes a protein that belongs to the ABC1 protein kinase family, suggesting that these two proteins may share a common function in the cell. The C. crescentus PepF protein possesses the conserved HEXGH motif present in zinc binding domains of...

Brisbarre, Nadège; Fardeau, Marie-laure; Cueff, Valerie; Cayol, Jean-luc; Barbier, Georges; Cilia, Valérie; Ravot, Gilles; Thomas, Pierre; Garcia, Jean-louis; Ollivier, Bernard.
A strictly anaerobic, slightly halophilic and moderately thermophilic, sporulating rod designated strain DVird3(T) was isolated from deep-sea hydrothermal vent samples collected at a depth of approximately 800 m on the Atlantic Ocean Ridge. Strain DVird3(T) possessed a few laterally inserted flagella, had a DNA G+C content of 33.1 mol% and grew optimally at pH 6.6 and at 45 degreesC. Growth was observed at temperatures between 20 and 58 degreesC and at pH values between 5.8 and 8.2. The optimum NaCl concentration for growth was 3% sea salt (30 g l(-1)); no growth was observed in the presence of 15 or 60 g sea salt l(-1). Strain DVird3(T) is heterotrophic and utilizes some sugars and various single amino acids. Acetate was the main fatty acid detected from...

Comparaison de methodes directes (microscopiques) et indirectes (par mise en culture) dans l'evaluation d'une pollution bacterienne d'origine fecale. Etude preliminaire ArchiMer
Munro, P; Bianchi, M.
Cells of two serotypes of Escherichia coli (O26B6 and 0111B4) were counted using epifluorescence and immunofluorescence microscopy (nonavalent serum) and plate count on specific medium. Values from indirect counts were 10 fold smaller than those from both microscopic counts. In a sample of sea water from an area receiving a fecal pollution, the highest number of bacteria was obtained by epifluorescence microscopy. In that case, the number of cells counted by immunofluorescence microscopy was 10 fold smaller than the number of bacteria able to grow on the medium. It could be due to: the diversity of the natural community, the possibility to other bacteria than E. coli to grow on the used culture medium, the restrictiveness of the serum for "pathogenic...

Comparison of commercially-available RNA extraction methods for effective bacterial RNA isolation from milk spiked samples Electron. J. Biotechnol.
García-Nogales,Paula; Serrano,Alicia; Secchi,Sonia; Gutiérrez,Serafín; Arís,Anna.
Nucleic-acid based methods for bacterial identification are extremely useful in diagnostic applications due to their specificity and sensitivity. However, they require an optimal purification of the target molecules. As part of the development of a new diagnostic method for the detection of bacterial RNA in cow milk, we have compared four commercially available RNA extraction kits for the isolation of bacterial RNA from spiked UHT milk samples. The kits were compared in terms of extraction efficiency and RNA purity using two bacterial species, the Gram negative Escherichia coli and the Gram positive Staphylococcus aureus. Two kits are based in silica-matrix extraction, and the other two in the guanidinium thiocyanate-phenol-chloroform extraction. In our...

Comparison of serum PCT and CRP levels in patients infected by different pathogenic microorganisms: a systematic review and meta-analysis BJMBR
Tang,Jun-Hua; Gao,Dong-Ping; Zou,Peng-Fei.
To avoid the abuse and misuse of antibiotics, procalcitonin (PCT) and C-reactive protein (CRP) have been used as new approaches to identify different types of infection. Multiple databases were adopted to search relevant studies, and the articles that satisfied the inclusion criteria were included. Meta-analyses were conducted with Review Manager 5.0, and to estimate the quality of each article, risk of bias was assessed. Eight articles satisfied the inclusion criteria. The concentrations of both PCT and CRP in patients with bacterial infection were higher than those with non-bacterial infection. Both PCT and CRP levels in patients with G− bacterial infection were higher than in those with G+ bacterial infection and fungus infection. In the G+ bacterial...

Conjunctival endogenous microbiota in patients submitted to cataract surgery BJM
Locatelli,Claudete I.; Kwitko,Sérgio; Simonetti,Amauri Braga.
Bacterial isolation, identification and antimicrobial susceptibility tests were carried out in ocular material collected with swab and polimethylmethacrylate (PMMA) or silicone intraocular lenses (IOL) from forty six patients submitted to cataract surgery. Seventy six isolates and seven different microorganisms were identified. Coagulase-negative staphylococci (CNS) were the predominant microorganisms isolated from swabs (71.4% of cases), PMMA lenses (81.3%) and silicon lenses (77.8%). Coagulase-negative staphylococci isolates revealed high resistance to penicillin G followed by tetracycline, chloramphenicol and aminoglicosides. However, these isolates displayed great susceptibility to vancomycin, cephalothin and ofloxacin. Except for penicillin G,...

Construction and use of a dialysis chamber for investigating in situ the toxicity of heavy metals on bacteria. ArchiMer
Tan, T.
Teflon dialysis sampling chambers have been employed to study the toxicity of ZnCl sub(2) on a cadmium-sensitive bacterium, Acinetobacter) CS 13, in an aquarium with running seawater. A concentration of 10 mg Zn super(++) l super(-1) did not influence the cell densities in the chambers, but cell densities decreased at zinc concentrations of 15 and 20 mg l super(-1). In seawater with 20 mg Zn super(++) lu-1 the bacteria accumulated zinc from 827 to 2813 mu g g super(-1) cells dry wt after 1 d. After 2 d the zinc concentration increased to 4625 mu g g super(-1) dry wt. Three chambers were filled with Acinetobacter CS 13 cells added to sterile sediment at a concentration of 1 x 10 super(8) cells ml super(-1) wet sediment. To test the security, these chambers...

Co-occurring nematodes and bacteria in submarine canyon sediments ArchiMer
Rzeznik-orignac, Jadwiga; Puisay, Antoine; Derelle, Evelyne; Peru, Erwan; Le Bris, Nadine; Galand, Pierre E..
In submarine canyon sediments, bacteria and nematodes dominate the benthic biomass and play a key role in nutrient cycling and energy transfer. The diversity of these communities remains, however, poorly studied. This work aims at describing the composition of bacteria and nematode communities in the Lacaze-Duthiers submarine canyon in the north-western Mediterranean Sea. We targeted three sediment depths for two consecutive years and investigated the communities using nuclear markers (18S rRNA and 16S rRNA genes). High throughput sequencing combined to maximal information coefficient (MIC) statistical analysis allowed us to identify, for the first time, at the same small scale, the community structures and the co-occurrence of nematodes and bacteria...

Copper II - polar amino acid complexes: toxicity to bacteria and larvae of Aedes aegypti Anais da ABC (AABC)
RODRIGUES,THIAGO A.D.; ARRUDA,EDUARDO J. DE; FERNANDES,MAGDA F.; CARVALHO,CLAUDIO T. DE; LIMA,ALESSANDRA R.; CABRINI,ISAÍAS.
ABSTRACT Control strategies using insecticides are sometimes ineffective due to the resistance of the insect vectors. In this scenario new products must be proposed for the control of insect vectors. The complexes L-aspartate Cu (II) and L-glutamate-Cu (II) complexes were synthesized and characterized by elemental analysis, visible ultraviolet, infrared spectroscopy and potentiometric titration. The toxicity of these complexes was analyzed in Aedes aegypti (Diptera: Culicidae) larvae and Gram-negative and Gram-positive bacteria. The interaction between the ligands and the amino acid balance and the distribution of the species as a function of pH were discussed. The lethal concentration median (LC50) for Ae. aegypti larvae were: L-glutamic acid-Cu (II) -...

Cultivation-Assisted Genome of Candidatus Fukatsuia symbiotica; the Enigmatic "X-Type" Symbiont of Aphids ArchiMer
Patel, Vilas; Chevignon, Germain; Manzano-marin, Alejandro; Brandt, Jayce W.; Strand, Michael R.; Russell, Jacob A.; Oliver, Kerry M..
Heritable symbionts are common in terrestrial arthropods and often provide beneficial services to hosts. Unlike obligate, nutritional symbionts that largely persist under strict host control within specialized host cells, heritable facultative symbionts exhibit large variation in within-host lifestyles and services rendered with many retaining the capacity to transition among roles. One enigmatic symbiont, Candidatus Fukatsuia symbiotica, frequently infects aphids with reported roles ranging from pathogen, defensive symbiont, mutualism exploiter, and nutritional co-obligate symbiont. Here, we used an in vitro culture-assisted protocol to sequence the genome of a facultative strain of Fukatsuia from pea aphids (Acyrthosiphon pisum). Phylogenetic and genomic...
